NTNG1 Modulates Progressions of Prostate Cancer Cells through JAK/STAT Signalling Pathway

چکیده
Netrin-G1 (NTNG1) is a glycosyl-phosphatidylinositol-affixed synaptic adhesion molecule that participates in carcinoma developments. However, its underlying mechanism regulating prostate cancer (PCa) uncertain. NTNG1 mRNA and protein expressions tissue samples cells were demonstrated to be promoted using RT-qPCR western blot. Thereafter, CCK-8, overexpression accelerated PCa cell viability while suppressed restrained viability. Additionally, transwell results indicated migratory invasive abilities of also facilitated by overexpressed but inhibited with suppression. Furthermore, RT-qPCR, Janus kinase 1 (JAK1) has been detected upregulated upregulation downregulation. Moreover, JAK1, JAK2, signal transducer activator transcription 3 (STAT3), Ki67 E-cadherin the knockdown elevated overexpression. In cells, acted as an oncogene through activating JAK/STAT3 signalling pathway.

CD147 modulates autophagy through the PI3K/Akt/mTOR pathway in human prostate cancer PC-3 cells
The multifunctional glycoprotein cluster of differentiation (CD)147 is highly expressed on the cell surface of the majority of cancer cells, and promotes tumor invasion, metastasis and growth. However, the role of CD147 in autophagy has not yet been explored in prostrate cancer cells. In the present study, prostate cancer PC-3 cells were cultured under starvation conditions, and the expression ...
متن کاملMicroRNA miR-125a-3p modulates molecular pathway of motility and migration in prostate cancer cells
Fyn kinase is implicated in prostate cancer. We illustrate the role of miR-125a-3p in cellular pathways accounted for motility and migration of prostate cancer cells, probably through its regulation on Fyn expression and Fyn-downstream proteins. Prostate cancer PC3 cells were transiently transfected with empty miR-Vec (control) or with miR-125a-3p. Overexpression of miR-125a-3p reduced migratio...
متن کاملβ1, 4-N-acetylgalactosaminyltransferase III modulates cancer stemness through EGFR signaling pathway in colon cancer cells
Cancer stem cells are cancer cells characterized with tumor initiating capacity. β1,4-N-acetylgalactosaminyltransferase III (B4GALNT3) synthesizes GalNAcβ1-4GlcNAc (LacdiNAc) which contributes to self-renewal of mouse embryonic stem cells. We previously showed that B4GALNT3 overexpression enhances colon cancer cell malignant phenotypes in vitro and in vivo. However, the role of B4GALNT3 in canc...
متن کاملRadiosensitizing effects of Sestrin2 in PC3 prostate cancer cells
Objective(s): The stress-responsive genes of Sestrin family are recognized as new tumor suppressor genes in breast carcinoma, however, the function of Sestrin family in human prostate cancer is not clear. Ionizing radiation (IR) is known to induce Sestrin gene expression in breast cancer cells. However, the response of Sestrin to IR has not been reported in PC3 prostate cancer cells. Materials ...
متن کاملMonepantel induces autophagy in human ovarian cancer cells through disruption of the mTOR/p70S6K signalling pathway.
We have recently shown that the novel anthelmintic drug monepantel (MPL) inhibits growth, proliferation and colony formation, arrests the cell cycle and induces cleavage of PARP-1 in ovarian cancer cell lines. Here we report on the mechanism behind the anticancer properties of MPL. The cytotoxic effect of MPL on ovarian cancer cells (OVCAR-3 and A2780) was investigated employing a panel of test...
